  • 方法一:以香草醛为原料,先用硫酸二甲酯甲基化得3,4-二甲氧基苯醛(藜芦醛),然后与甲氧基丙腈缩合生成3',4'-二甲氧基-2-氰基-3-甲氧基丙烯,再经加成,最后与硝酸胍环合而得。 香草醛溶于氢氧化钠溶液于60-80℃滴加硫酸二甲酯,并于碱性条件(HP7-9)下回流反应3h。反应液稍冷后用甲苯萃取,萃取液回收溶剂后得藜芦醛。在甲醇与甲醇钠溶液中,藜芦醛与甲氧基丙腈于65-70℃下回流反应5h。加水冷却后结晶,过滤、干燥得3',4'-二甲氧基-2-氰基-3-甲氧基丙烯。 上述缩合物在甲醇、甲醇钠中与硝酸胍于75℃下搅拌反应2h,然后于95℃下继续反应5h。冷却后析出结晶,过滤得二甲氧苄氨嘧啶粗品。粗品经乙酸重结晶、活性炭脱色(95℃,1h),氨水碱析、分离、干燥得成品。
