  • 方法一:洛克沙生的合成路线较多,主要有:①以对羟基苯胂酸为原料经混酸在低温下消化而得;②3-硝基-4-羟基苯胺经重氮化、置换(胂化)而得得;③以苯酚为原料经胂化、硝化而得;④对氨基苯胂酸经重氮化置换得羟基苯胂酸,再硝化而得;⑤以对羟基苯胺为原料经重氮化、胂化、硝化制得。从原料来源、成本高低、路线长短、收率高低和操作难易等衡量,路线③和⑤具有优势,适合于工业化生产。 合成⑤的反应式和步骤如下: 将109.5对氨基苯酚和工业盐酸135.5g加入反应器,于室温下搅拌,使其充分成盐。冷却至0℃,滴加30%的亚硝酸钠溶液反应生成重氮盐溶液。控制温度不超过5℃。以淀粉-KI试纸检查重氮化终点。 将98.9g A2O3和30%的碳酸钠溶液在反应器中升温搅拌,使其完全溶解,并沸腾0.5小时。冷却至10℃,加入几滴CuSO4溶液,将重氮盐溶液缓缓加入此含有盐酸铜的亚砷酸钠溶液,反应生成对羟基苯胂钠溶液,控制温度不超过10℃。加稀盐酸酸化,过滤,滤液中和至中性,真空浓缩,再加少量活性炭脱色,趁热过滤,滤液放置过夜,析出结晶,过滤得粗品。粗品溶于水,稀盐酸酸化,冷却至10℃,放置过夜重结晶,经过滤、干燥得到对羟基苯胂酸171.8g,熔点172-174℃(分解),收率78.1%。 将110g对羟基苯胂酸搅拌于397mL硫酸中,降温至0℃后滴加混酸(23%硫酸、44%硝酸,其余为水)150.9g,控制温度不超过18℃,加完后维持20℃反应2h。将反应物倾倒入冰水中,过滤并水洗至中性,粗品加去离子水溶液,加入少量活性炭加热脱色,趁热过滤。滤液放置2天,析出结晶,经过滤、真空干燥得102.9g产品,熔点300-302℃,收率78.2%。 另外,也可以以氯苯胺为原料合成。有关 将对氯苯胺12.8g(0.1mol)溶于60mL水、18mL浓盐酸溶液中,于0-5℃下慢慢滴加亚硝酸钠6.9g(0.1mol)和25mL水的溶液。滴加完毕后保温加入氯化亚铜1.5g和有机硅消泡剂1Ml,再慢慢滴加由19.8g(0.1mol)三氧化2砷和8g(0.2mol氢氧化钠溶于100mL水组成的溶液。滴加完毕后再加入10%的氢氧化钠溶液10mL,继续反应1h,然后升温至60℃反应1h。抽滤,滤液加浓盐酸调PH=4,再抽滤,滤液浓缩至约80mL,加活性炭脱色过滤。滤液再加浓盐酸调PH=2,置于冰箱过夜,收集沉淀,水洗后干燥得20.4g微黄色晶体对氯苯胂酸,收率86.3%,熔点大于270℃。 将硝酸钠12.5g(0.15mol)溶于60mL浓硫酸中,搅拌下慢慢滴加对氯苯胂酸23.6g(0.1mol)。升温至80-85℃,反应2h后结束。冷却至室温,慢慢倾入100g冰和100mL水的混合物中。过滤、水洗、干燥得24.5g淡黄色结晶体4-氯-3-硝基苯胂酸,收率87.2%,熔点大于270℃。 将氢氧化钠15.0g(0.375mol)溶于35mL水,再加入4-氯-3-硝基苯胂酸14.1g(0.05mol),于95-100℃下搅拌反应10h。然后加入30mL水,用浓盐酸调PH=10,加入活性炭于80℃下脱色过滤。滤液再加浓盐酸调PH=2,置冰箱中过夜。过滤、水洗、干燥得9.4g黄色结晶,熔点大于270℃,收率71.5%。
