  • 嘌呤代谢:人体中的嘌呤来自两方面:即食物中所含的核酸和由机体合成。构成核酸的嘌呤核苷酸主要为鸟嘌呤核苷酸和腺嘌呤核苷酸。其主要合成途径如下:先由核糖与磷酸合成5-磷酸核糖,与三磷酸腺苷作用,生成1-焦磷酸-5-磷酸核糖(PRPP),后者与谷氨酰胺作用,在磷酸核糖焦磷酸酰氨移换酶的催化下,生成1-氨基-5-磷酸核糖(PRA),然后在一系列酶的催化下通过十步反应生成次黄嘌呤核苷酸,再由次黄嘌呤核苷酸通过两步反应分别生成鸟嘌呤核苷酸和腺嘌呤核苷酸,并用以合成核酸。嘌呤核苷酸的主要分解途径为:次黄嘌呤核苷酸、鸟嘌呤核苷酸和腺嘌呤核苷酸,在核苷酸酶的催化下分别生成各自的嘌呤核苷,再在核苷磷酸化酶的催化下分别转变为次黄嘌呤、鸟嘌呤和腺嘌呤。鸟嘌呤在鸟嘌呤酶催化下可生成黄嘌呤;但腺嘌呤由于人体缺乏腺嘌呤脱氨酶,故只能在腺嘌呤核苷或腺嘌呤核苷酸水平上分别受腺嘌呤核苷脱氨酶或腺嘌呤核苷酸脱氨酶作用而生成次黄嘌呤核苷或次黄嘌呤核苷酸。次黄嘌呤在黄嘌呤氧化酶催化下氧化为黄嘌呤,黄嘌呤为黄嘌呤氧化酶催化而氧化为尿酸,后者为嘌呤核苷酸分解代谢的最终产物,随尿排出体外。嘌呤的合成代谢受酶促产物的反馈调节。1-焦磷酸-5-磷酸核糖与谷氨酰胺作用,为磷酸核糖焦磷酸酰氨移换酶所催化而生成1-氨基-5-磷酸核糖,是重要的首步反应。腺嘌呤核苷酸、鸟嘌呤核苷酸对磷酸核糖焦磷酸酰氨移换酶有抑制作用,是负反馈调节点之一;而次黄嘌呤核苷酸在转变为鸟嘌呤核苷酸和腺嘌呤核苷酸的过程中,亦受后二者的负反馈调节。故嘌呤核苷酸合成与分解代谢,得以维持动态平衡。此外,某些抗代谢物对于嘌呤核苷酸的合成代谢,也有抑制作用。上述反应的主要过程及其调节见图3所示。 图3为嘌呤的合成和分解代谢途径及合成反馈调节。 E1磷酸核糖焦磷酸酰胺移换酶;E2次黄嘌呤核苷-5′-磷酸脱氢酶;E3鸟嘌呤核苷-5′-磷酸脱氢酶;E4腺苷酸代琥珀酸合成酶;E5腺苷酸代琥珀酸酶;E6腺嘌呤磷酸核糖转移酶;E7次黄嘌呤-鸟嘌呤磷酸核糖转移酶;E3腺嘌呤核苷脱氨酶;E9核苷磷酸化酶;E10鸟嘌呤酶;E11黄嘌呤氧化酶;→主要合成途径;→补救合成途径;→合成反馈抑制;→分解途径。

  • 嘌呤、嘧啶等物质的作用:嘌呤和嘧啶是构成细胞中核糖核酸和脱氧核糖核酸的重要物质。大麦和麦芽含有0.2%~0.3%的核酸干物质,在糖化时,受各种磷酸酯酶的降解而形成核苷酸、核苷、嘌呤和嘧啶等多种降解物,其中只有嘌呤和嘧啶进入酵母细胞,构成核糖核酸、脱氧核糖核酸、三磷酸腺苷和某些辅酶,核苷酸很难被吸收。若培养基中缺乏嘌呤和嘧啶,就得依靠碳水化合物和氨基酸去合成,这样就要消耗很多能量和影响酵母的增殖。一般说,麦汁中不会缺乏需要的嘌呤和嘧啶。 嘌呤、嘧啶代谢障碍:嘌呤和嘧啶是生物(包括人类)在核酸代谢中所必需的杂环含氮化合物。嘌呤、嘧啶与核糖及磷酸盐结合形成RNA;嘌呤、嘧啶与去氧核糖及磷酸盐结合产生DNA。DNA是组成基因的主要化学成分,对基因的传递即遗传功能起重要作用;RNA的主要作用为调节细胞内蛋白质的合成。嘌呤代谢的最终产物主要为尿酸。与临床有关的嘌呤类有腺嘌呤及鸟嘌呤。重要的嘧啶有胸腺嘧啶、胞嘧啶和尿嘧啶。在嘌呤及嘧啶代谢中必须有各种酶参与才能使代谢过程按正常步骤进行,缺乏其中任何一种酶均可出现相应的嘌呤或嘧啶代谢障碍,以致出现临床特异性症状,如乳清酸尿症、黄嘌呤尿症及Lesch-Nyhan综合征. 其他如原发性儿童痛风;尿酸血症伴部分磷酸核糖转移酶缺陷;女性智能迟缓、自身突变及高尿酸血症综合征;2,8-去羟腺嘌呤尿症及去氨酶缺陷等在儿童少见。 参考资料:管敦仪 编著.啤酒工业手册.北京:中国轻工业出版社.1998.第366页.
